The purification step (RP-HPLC) is one of the main bottlenecks in the peptide synthesis workflow, limited by small loading amounts, long separation times, poor recoveries and high costs. Biotage® Sfär Bio cartridges offer similar performance, in a flash format, at a fraction of the cost. They have an increased loading capability compared to prep RP-HPLC, enabling more peptide to be processed in a single injection and faster, more efficient peptide purification.

This work demonstrates that peptide purification efficiency by flash

In this study we synthesized two closely related amphipathic peptides and show the mass directed flash purification of non-acetylated ‘18A’ from a crude mixture containing a single residue deletion sequence ‘17A’ (-Glu residue) using the Biotage® Isolera Dalton™.

Improvements in solid phase peptide synthesis strategies and
development of resin linkages susceptible to low acid cleavage
conditions has enabled synthesis of long peptides while keeping the
protecting groups intact. This strategy is now used for the
preparation of chemically synthesized proteins, wherein shorter
peptide fragments are ligated together. They are also found in the
synthesis of peptide macrocycles that utilize head-to-tail cyclization strategies. Although linear synthesis of protected peptides is generally straightforward, purification of these compounds using traditional reversed phase methods is quite challenging. Herein we describe the use of normal phase chromatography for purification of fully protected peptides.

Although capable of very high resolution, RP-HPLC is often limited by low column loading capacity, therefore demanding a significant time investment for peptide purification. As an alternative strategy, reversed-phase flash chromatography can also be used to purify synthetic peptides. The larger particle size used in flash column chromatography enables much larger loading capacity, thereby significantly reducing the time required for peptide purification.

Peptide purification via flash
chromatography has recently been demonstrated as a viable
alternative to the more standard HPLC methods currently
utilized. Flash chromatography offers peptide chemists the
advantage of significantly greater loading capacity reducing the
overall purification time but with a compromise of decreased
peak resolution. Herein we present several strategies that,
when implemented, allow for very high purity peptide samples
purified by flash chromatography.
