This application note describes a procedure for sample
pre-treatment and extraction of a panel of 49 drugs of
abuse from human hair, using Biotage® Lysera for matrix
micropulverization, prior to clean up using ISOLUTE® SLE
supported liquid extraction. The application note contains
procedures optimized for 400 μL capacity column format and
200 μL and 400 μL supported liquid extraction plate formats,
for higher throughput applications. The methodology delivers
clean extracts and analyte recoveries mostly greater than
80% with RSDs lower than 10% for all analytes and LLOQ from
1 ng/mL.

New, novel benzodiazepines have become some of the most abused unique psychoactive substances seen today. In the United States, these compounds are not approved for prescription use. Therefore, the demand for testing of these drugs has rapidly increased, both in the United States and worldwide. Urine and whole blood are common matrices of choice for forensic laboratories as they can be easy to collect, and they provide relevant information regarding recent or active use of illicit materials.

Hydroxychloroquine (HCQ) and chloroquine (CQ) are common antimalarial drugs with long half-lives. They are currently being tested as potential treatments for COVID-19. Due to their high publicity in the media, many forensic toxicologists are working on developing methods to extract and detect these compounds in different matrices to understand the adverse effects, potential toxicity, and drug interactions associated with these drugs. These compounds are not typically included in general unknown screens. A simple sample preparation procedure that delivers clean extracts and analyte recovery greater than 90% for CQ and greater than 82% for HCQ with minimum matrix effects and high process efficiency was developed.

LC-MS/MS analysis of volatile analytes is problematic when 96-well plates are used. Solid phase extraction of urine with high drug concentrations can cause contamination of the elution solvent during evaporation as the compounds volatilize then redistribute in adjacent wells.This phenomenon, known as “evaporative crosstalk”, can generate false positive results.

Evaporative crosstalk is well-to-well cross contamination
during extract evaporation after SLE or SPE extraction in a 96
well plate. It is often observed with volatile analytes like
methamphetamine and amphetamine, but it can also occur
with other drugs and metabolites when urine samples have

Biotage® SPE Dry is designed with novel technology and a simple interface to produce uniform and reproducable evaporation. The design has a small footprint to fit easily in the fume hood. The heating mechanism is designed to rapidly heat without overshooting the temperature, preventing degradation of temperature sensitive compounds. Standard SPE Dry 96 and SPE Dry 96 Dual systems are supplied with stainless steel needles. For high purity work, or applications that require particularly aggressive solvents, PTFE coated needles are available. Both needle types can be individually replaced should they become damaged or corroded. SPE Dry is available in single or dual format and adapts easily to 24-, 48-, 96-, and 384-well microplate formats.
Efficient solvent evaporation is facilitated by heated gas flow from above and below the plate(s). Optimum temperature and gas flow conditions are set by the user. Evaporation conditions for the upper and lower heads can be controlled independently.
These easy to use systems have simple front panel controls for regulating gas temperature and flow rate. Valuable fume hood space is saved with the compact design.
The removable heads are easy to clean and can be sonicated as a whole unit. Removable needles are easy to replace.
SPE Dry 96 accommodates one microplate, while SPE Dry 96 Dual is a two-microplate system. The standard systems are supplied with stainless steel needles and are compatible with 24 and 96-well microplates. Higher density arrangements are also available for 384-well microplates. PTFE coated needle assemblies are available for use with aggressive solvents.
