This application note describes the extraction of a panel of estrogenic hormones from human serum using EVOLUTE® EXPRESS ABN solid phase extraction plates prior to LC/MS analysis. The simple sample preparation procedure delivers clean extracts and analyte recoveries greater than 90% with RSDs lower than 5% for all analytes. Linearity of greater than 0.99 is achieved for Estrone (E1) and Estradiol (E2) from 5–2000 pg/mL, and 50–20000 pg/mL for Estriol (E3). No derivatization is required, and detection limits are enhanced using a fluorinated mobile phase.

Analysis of drug panels in urine samples can be challenging, and the trend towards larger panels including multiple drug classes compounds the issues faced during method development.
This white paper examines a number of aspects of sample preparation, and their impact on the success of subsequent LC-MS/MS analysis of broad urine panels.
Section 1 examines the applicability of various sample preparation techniques: supported liquid extraction, reverse phase SPE and mixed-mode SPE, to the various classes of drugs extracted. In addition, hydrolysis approaches: enzyme type and protocol used (time, temperature), are compared.
Mixed-mode reverse phase/cation exchange SPE is widely used for extraction of basic drug classes from urine, but the inclusion of drugs and metabolites that exhibit ‘non-typical’ functionality within urine panels can be problematic. Section 2 examines the impact of various parameters (interference wash strength, elution solvent composition) on analyte retention, elution and extract cleanliness with particular focus on zwitterionic (gabapentin, pregabalin) and non-ionic (carisoprodol, meprobamate) drugs.

This application note describes the extraction of a panel of 19 steroid hormones from human urine using EVOLUTE® EXPRESS ABN solid phase extraction plates prior to LC-MS/MS analysis. The simple sample preparation procedure delivers clean extracts and analyte recoveries greater than 75% with RSDs lower than 5% for most analytes. Linearity of greater than 0.999 is achieved for all analytes in the range 1–1000 pg/mL.

This application note describes the fully automated extraction of carboxy-THC from urine, following base hydrolysis prior to GC/MS analysis. The method was automated using Biotage® ExtraheraTM, configured for use with EVOLUTE EXPRESS ABN columns.
The method delivers clean extracts and analyte recovery of 90% with RSD lower than 10% for carboxy-THC and its deuterated internal standard. Using the Biotage® ExtraheraTM, 24 samples were extracted in approximately 38 minutes. The limit of quantitation of 2 ng/mL is below the SAMHSA/ EWDTS confirmation cut off of 15 ng/mL for workplace testing applications.

Particular emphasis was placed on the sample preparation to

Analysis of drug panels in urine samples can be challenging, and the trend towards larger panels including multiple drug classes compounds the issues faced during method development.
This white paper examines a number of aspects of sample preparation, and their impact on the success of subsequent LC-MS/MS analysis of broad urine panels.
Section 1 examines the applicability of various sample preparation techniques: supported liquid extraction, reverse phase SPE and mixed-mode SPE, to the various classes of drugs extracted. In addition, hydrolysis approaches: enzyme type and protocol used (time, temperature), are compared.
Mixed-mode reverse phase/cation exchange SPE is widely used for extraction of basic drug classes from urine, but the inclusion of drugs and metabolites that exhibit ‘non-typical’ functionality within urine panels can be problematic. Section 2 examines the impact of various parameters (interference wash strength, elution solvent composition) on analyte retention, elution and extract cleanliness with particular focus on zwitterionic (gabapentin, pregabalin) and non-ionic (carisoprodol, meprobamate) drugs.

There is no fixed expiry date for these products.
All ISOLUTE and EVOLUTE products are packaged in inner metal laminate barrier bags, which provide an effective barrier to oxygen and moisture penetration. Recommended storage conditions are standard (see below).

It's unique modified surface structure is water-wettable to promote robust, reliable sample preparation. A balanced combination of polar (hydrophilic) and non-polar (hydrophobic) interactions allows efficient extraction of analytes of wide ranging polarity. EVOLUTE® ABN sorbent is available in EVOLUTE® EXPRESS 96-well plate and column formats for use with fast Load-Wash-Elute procedures. Eliminate traditional conditioning and equilibration and increase throughput without compromising performance.
EVOLUTE® sorbents have a narrow pore diameter, tailored to eliminate co-extraction of proteins from biofluids. Removal of proteins during sample preparation avoids transfer in to the analytical system, reducing matrix effects in LC-MS/MS, and avoiding contamination of the analytical column. This minimises the need for frequent backflushing and guard columns replacement.
EVOLUTE® ABN uses a single generic procedure suitable for a wide range of analytes in aqueous matrices. Without secondary interactions, pure organic elution solvents can be used, reducing evaporation time and providing optimal LC-MS/MS compatibility, making the transfer to analysis more efficient. EVOLUTE ABN’s narrow particle size distribution provides consistent flow characteristics for a variety of sample matrices. Optimized packing techniques eliminate channelling, which can lead to analyte breakthrough.
Even viscous biofluids can be loaded onto EVOLUTE EXPRESS ABN products without the need for conditioning or equilibration, using fast load-wash-elute SPE.
