The extraction of basic drugs from biological fluids using a purely non-polar retention mechanism (e.g. C4, C8 or C18) can lead to extracts that contain a large amount of non-polar co-extracted material that can interfere with subsequent analysis. Conversely, extraction mechanisms based on ion exchange interactions can be non-robust due to the variable ionic strength of the sample matrix.

The first decision that needs to be made is what the final analysis will be for the analyte. This will have an impact on the sample and cartridge size, as well as the final elution solvent. Gas chromatography offers higher sensitivity than HPLC, while HPLC is better suited for ionisable species and very high molecular weights. If LC-MS is available, minimal sample clean-up may be required.

What is the maximum capacity of an SPE column?
Answer:
The capacity of a sorbent is defined as the total mass of strongly retained analyte that can be retained by a given mass of the sorbent under optimum conditions.
For most sorbents, this can range from 1-5% by weight. 100 mg of sorbent might retain 1-5 mg of analyte.
Ion exchange sorbents typically have a capacity of 0.3-1 meq/g.

Analytes are initially retained by a non-polar retention mechanism, with inorganic salts, polar interferences such as phospholipids and large molecular weight peptides and proteins passing directly through the sorbent bed. The pH is then adjusted to allow an ion exchange retention mechanism to retain the analyte(s). Non-polar interferences are removed using an organic solvent or organic solvent/water wash. Analytes are then eluted in an extra clean fraction.
A good alternative to ISOLUTE HCX for basic compounds that requiremore retentive character from the mixed-mode sorbent.
ISOLUTE HCX-3 is available in a wide range of SPE columns and 96-well plate formats.
