Isolera™ is a generation of high performance purification systems. The family includes Isolera One, also available as Isolera Dalton 2000, Isolera Prime and Isolera LS. Isolera One and Isolera LS can be equipped with Spektra to become Isolera Spektra One and Isolera Spektra LS.
Spektra contributes to make Isolera a world leading automated flash system designed to purify synthesis reaction mixtures, natural product extracts, and other organic compound mixtures from milligram to multi-gram quantities. It features an additional host of both labour and cost saving features such as TLC-to-step gradient, full spectral analysis, lambda all collection, 3D view and improved baseline correction algorithms, allowing the same chemistries and purifications to be run but significantly reducing the amount of solvent required and time to do so. The systems are available in a number of different configurations and are so intelligent they will also recommend the optimum cartridge size for your sample.
Isolera™ Spektra One
Is a single cartridge flash purification system just right for use by one or two chemists who need to perform 1–5 purifications per day. Isolera Spektra One is compatible with all Biotage flash cartridges Biotage®, Sfär.
Isolera™ Spektra LS
Is the definitive flash chromatography system for reliable scaling up from grams to to multi, even hundred gram scale purification, using the common Isolera interface. Enjoy all of the sophisticated software features of the laboratory scale systems in a robust instrument built for scale-up labs. Isolera Spektra LS adds an integrated pump assisted loading mechanism for larger samples, and a funnel rack collection option which allows samples to be collected in larger fraction collection vessels.
Fast and Efficient
With running costs and time a concern in most labs, Isolera™ systems recommend cartridges based on the sample size and your TLC data, so the smallest column can be choosed, thus reducing costs and solvent consumption. They also work out the best solvent gradient based on compound, and provide real time indication of eluting compounds, using UV, ELSD or Mass Detection. They subtract baseline signals automatically, so chemists can be confident that the correct fractions are collected.

Isolera Assist offers two experience levels – for confident users, the advanced user profile opens up all of the functionality of the Isolera system. Alternatively, users can opt for a basic profile, where a streamlined interface ensures the user is up and running in the minimum time. User levels also assist in optimizing open access environments, where the two user levels operate side by side on the system.
Like the Isolera just as it is? If you are already very familiar with the Isolera, just use the ‘Chemistry’ menu button to use the classic Isolera interface.

Achieve up to 30% solvent savings with Gradient Optimization “GO”
Step-gradients can be short and powerful providing separations for one or more compounds but can be challenging to develop and optimize. Isolera Spektra provides gradient optimization through new, patent-pending TLC-to-Step Gradient technology. Using solvent and TLC Rf values, Isolera Spektra builds a gradient to separate all the compounds (up to 6) in the sample. This new technology will also provide cartridge selection guidance based on the cartridge loading capacity and purification speed. The step gradient can also be used to isolate a targeted compound reducing run time and solvent use further.
Advanced λ-All detection
Many compounds being purified have unknown absorbance spectra. The Isolera™ Spektra λ-All function records all available wavelengths for compound detection and sums the responses to maximize sensitivity and minimize sample loss.
Digging Deeper With PDA Spectral Analysis
Isolera Spektra brings Photodiode Array (PDA) detection to flash chromatography for the first time. The full spectrum for each compound can be seen as it elutes from the cartridge – in
real-time. This information can be used to confirm purity and compound identity. All spectra are stored and can be reviewed in 2D to verify fraction purity. Post fraction thin layer chromatography (TLC) to determine which fractions contain pure compound can be eliminated.
Many chromatographic solvents absorb UV light. Isolera Spektra uses advanced real gradient zeroing for baseline correction to eliminate background shifts. Using all wavelengths for detection with a gradient can lead to a rising baseline which will interfere with compound detection and cause inaccurate fraction collection.
3D graphics for the complete picture
Identify impurities by taking advantage of the PDA 3D chromatogram display. Chemists can view the chromatogram in terms of both elution volume and UV absorbance which provides even more purity confirmation.
Elute complex samples with the Quatro-binary gradient
Use up to four solvents in a single gradient to easily purify samples with diverse polarity. With Quatro-binary gradient capability traditional binary gradients with a limited polarity range can be adjusted to elute very lipophilic and highly polar compounds within a single purification. Or compound solubility during the separation by adding a constant amount of a co-solvent, acid, or base with the Isolera's advanced pump (not available on Prime).
Maintain compound solubility with a ternary co-solvent
Isocratically pump a third solvent into any binary gradient to help maintain compound solubility eliminating potential over-pressure issues caused by precipitating compounds.
Achieve superior purification with Biotage flash cartridges
The Isolera™ Spektra One systems can use various styles including Biotage Sfär and Biotage® SNAP with silica masses from 5 to 750 g.
